Document Summary

Many technologies rely on enzymes: enzymes used in genetic tests, analyses, engineering, etc. Rely on complementarity: looking for dna or rna that is complimentary to it. Most experiments start with a sample: work with dna in the sample first, run dna on a gel. First step is to produce enough dna to work with. Can utilize bacteria: bacteria has own nuclear genome, has ability to carry small dna plasmid molecules, bacteria can replicate the plasmid and divide. In bacteria, they are involved in eliminating dna from foreign sources. Two restriction enzymes cut phosphate backbone in different locations: 5" o(cid:396) 3" o(cid:448)e(cid:396)ha(cid:374)g, overhangs can be used to put things back together (cid:862) ti(cid:272)ky e(cid:374)ds(cid:863, complementarity of dna. 2 pieces of dna cut with the same re can be pasted back together: ligase seals nick. There may be: plasmid with dna of interest, plasmid with random dna, plasmid ligated on its own, plasmid that does not get into bacteria at all.
