Document Summary

Holds 5ml of mitochondria in suspension aka in buffer. Sealed airtight: atmospheric oxygen can"t get in. Oxygen electrode at bottom of this chamber measure oxygen concentration in that 5ml. (measure the rate of oxygen being evolved aka made) Got to kill some cells to purify mitochondria. Grind up chlamydomonas cells (lots of broken mitochondria, chloroplasts and nuclei but you can use special treatment to get pure sample of intact mitochondria) . Seal the chamber and then add molecules to the 5ml solution using a syringe. Introduce compound and see its effects on oxygen in the chamber (effects on mitochondrial respiration) Change in respiration rate (oxygen consumption) in isolated mitochondria by addition of nadh, adp, uncoupler etc: measure oxygen level before the addition of any mitochondria yet (0-2min). Obviously, there will be no change to oxygen levels. You haven"t added any mitochondria yet, they"re still in a buffer: adding mitochondria does not change the oxygen levels.
