Bacterial transcription initiates the promoter sequence: sequences at -10 and -35 important to promoter activity, -10 sequences are very rich in a t bases. 10 = 10 bases back from start of transcription. Promoters are not always very efficient (limit gene expression for certain genes) Strand used for template dependent on where promoter is located. There is no template strand for the whole genome: each gene could be on the bottom or top for the template.
