Polarity: essential in gene structure and gene expression. Mrna has a great deal of secondary structure: always pairs with itself not a ribbon shape trna base pairs with itself rrna base pairs with itself and is catalytic, ribozyme activity. Dna polymerase can make more copies of itself. Not made of bases no complementary base pairing. Organelles have typical bacterial gene structure and expression: mitochondria and chloroplast bacterial gene expression, nucleus - eukaryotic gene expression. Rna sequence binds to promoter and transcription begins. Bacterial transcription initiates the promoter sequence: sequences at -10 and -35 important to promoter activity, -10 sequences are very rich in a t bases. 10 = 10 bases back from start of transcription. Promoters are not always very efficient (limit gene expression for certain genes) Strand used for template dependent on where promoter is located. There is no template strand for the whole genome: each gene could be on the bottom or top for the template.
