If we wanted to look at wbc we would look at transmembrane proteins that are specifically produced from different cells. We have an antibody that recognizes thy1. 2 and a different antibody for cd3. Conjugate antibodies with green or red fluorophore and cell surface only recognize proteins outside the cell. Can use a permeable dye that recognizes inter cellular structure. 3 different types of fluorescence perimeters we can monitor. Thy 1 expressing cells will have more green. Cd3 expressing cells will have more red. This allows us to quantify the cells we labelled. Lasers focus on a fluid transporting tube. Fluid has labelled cells and travels through the tubes and the tubes get smaller and smaller as fluid goes through allowing individual cells to go through. As cells go through laser light can illuminate each cell. Light emitted is detected by light detectors a red and green light detector.
