Document Summary

Experiment 1: cellular origin of ig (ab)----- b cells make antibodies after establishing that t + b cells seem to. Experiment occurred before we knew about the ig loci. What was known: we have antibodies that recognize markers that distinguish b cells and t cells. T cells (were known as thy-1 or theta (cid:272)ells(cid:895): th(cid:455)(cid:373)o(cid:272)(cid:455)tes (cid:449)e(cid:396)e used as a sou(cid:396)(cid:272)e of (cid:858)t (cid:272)ells(cid:859) B cells have membrane bound immunoglobulin, (cid:271)o(cid:374)e (cid:373)a(cid:396)(cid:396)o(cid:449) used as sou(cid:396)(cid:272)e of (cid:858)b (cid:272)ells(cid:859) What (cid:449)as(cid:374)(cid:859)t k(cid:374)o(cid:449)(cid:374): (cid:449)hich cell is producing antibodies. System: isolate thymocytes (t cell source) from balb/c mouse, mix them with bone marrow cells (source of b cells) from a balb/c mouse. Lethally irradiate another balb/c mouse (recipient)- this mouse will have no cells which allows us to only see responses from thymocytes, bm cells, antigen and antibody. Take solution of antigen, bm cells, thymocytes and inject it into the irradiated recipient balb/c mouse. Have to make sure the cells are histocompatible.
