What is microbiology: the scientific study of microorganisms and their effects. Microorganisms are organisms invisible to the naked eye. Magnification and resolving power of microscopes: you cannot see objects that are less than 100 micrometer. Resolving power: the resolving power of an optical systems is its ability to distinguish - form a clear image - between small objects in close proximity, our eyes can"t form images of microbes: Our eyes resolving power is 0. 1 mm (100. 100 m cannot be clearly distinguished by our eye. In addition, we have to consider the limit of light diffraction: na is related with the amount of light that the lens can gather. Why do we use immersion oil: to increase resolving power of microscope; immerse objective lens and specimen in oil (with high refractive index) so that the numerical aperture of the objective lens increases. Why shortest wavelength: used to generate image, the lower the bending will be will lead to good image.
