Document Summary

Semiconservative replication new dna duplex made of one parental and one new strand. Conservative replication new dna duplex consists of two newly synthesized daughter strands parental duplex intact. Both strands are labeled with heavy nitrogen (15n). Unwinding of the dna duplex results in a replication fork occurs in the 5" to 3" direction. Rna primase (an rna polymerase) lays down an rna primer. Dna polymerase extends the rna primer. cannot begin a new strand on its own, only adds to existing nucleotides. Primers removed and replaced with dna, fragments of the discontinuous (lagging) strand are ligated. Dna polymerase removes the primer and replaces it with dna. Dna ligase forms a bond joining okazaki fragments. Topoisomeras e ii relieves the stress of unwinding. Single-stranded binding proteins stabilize single strands of dna. Dna polymerase complexes of each strand stay in contact build strands at same rate. Requires lagging strand be looped around to allow contact between the polymerase complexes to occur.
