G tells us about reaction equilibria , while g tells us about reaction rates . G can"t be used to access kinetics of a reaction. By lowering g via catalysis, we can increase the reaction rate. We can"t measure g , but we can measure reaction rate ! (by measuring how much products is formed compared to the reaction time). Michaelis-menten equation : provided a way to mathematically analyze enzyme catalysed reactions. Subtilisin : extracellular proteolytic enzyme synthesized by gm +ve bacteria (e. g. bacillus subtilis ) Problem breaks peptide bonds via nucleophilic attack commercially used as a stain cutter inactivated by peroxides/bleach found in laundry detergent this is due to oxidation of an important active site residue, met-222 & the formation of methionine sulfoxide. Solution : replace met-222 with another residue that does not react with peroxide yet still allows the enzyme to function. We won"t know which a. a. is better until we run an experiment.
