The cargo receptors extend through the membrane and interact with the cargo molecules in the cytoplasm. As the process continues, the vesicles round up and pinch off. By accumulation of proteins a piece of the membrane can separate in a circle. The proteins orientation with respect to the cytoplasm and the interior membrane-bound compartments is maintained during its travel through the endomembrane system.
