Cytokines low mw reg proteins/glycoproteins secreted by wbc, mon, macro, ect: interleukins: il-1 to il-35, common names: tumour necrosis factor (tnf), inteferons (ifn, chemokines (attract immune cells to inflame/infection) High affinity for rs so only need a little (nanomolar), act locally. Intra signaling cascade are act when cytokines bind specific rs. Cytokine binds alpha r cytoplasmic chain which recruits b chain (dimerize) Jak kinases stuck to chains p eachother + add p to cytoplasmic tail to recruit stat. Stats form dimer and translocate to nucleus + initiate gene transc: pleitropy: can act on diff cells, ex. Pro-inflam made mostly by macro, dc + other. Inflame , hematopoiesis, cellular prolif and differentiation and wound healing. Cytokine storm: too many pro cytokines " inflame disease, tissue damage, organ failure ex. Made mostly by macro + t cells, limit inflame response by inhibiting pro-inflam prod and act immune cells for healing. Too much anti= lack of immune response too little= autoimmune disease.
