Drugs that are non-polar diffuse across plasma membranes and are absorbed. Drugs with this characteristic are also easily distributed through body tissues, but only free drug molecules can be distributed. Fat soluble state in the blood will go to the target tissue. The charged form of the drug will slowly become fat soble in the blood and can get into the target tissue. Only free (unbound) drug molecules can be distributed into body tissues. Blood flow, organ size and physical properties (solubility) of the drug dictate distribution. Aspirin and warfarin should not be taken together, since aspirin kicks warfarin off of albumin = risk of hemorrhage. The more blood flow a organ has, more drug it will get. P-gp present in the intestine, blood-brain barrier, fetal-placenta barrier unless the drug is highly fat soluble. Parenteral administration : drugs distribute to target tissues and exert a biological/therapeutic effect.
