Mimic normal er behaviour contain all of the same membrane-associated proteins: microsome membranes treated with detergent releases proteins inside microsomes. Added protease digests the proteins: right: membrane left intact and protease is added, newly synthesized proteins are protected from proteases within, newly synthesized proteins move directly into microsomes. Hypothesis: if translation is completed, transport cannot. Control: proteins synthesized in vitro in the presence of occur microsomes: mature proteins found inside microsomes.
