- When B cell receives cytokine signal, transcription is activated upstream of constant region
o This does not result in productive mRNA
- DNA is now accessible to AID
o Because it has opened up for transcription

- Nicks are made on both DNA strands
- Double strand breaks in DNA upstream of constant region to be recombined
- DS break repair machinery repairs break by cutting out the intervening DNA
- Selected region now adjacent to VDJ region
- Switch regions
o Low transcription from this
o No functional mRNA
