How does this work: interested in a specific process and want to know more about how it works. Investigation of a biological process using mutants: identify genes and proteins that act in process. Genetic screens: mutant hunt, look through natural or mutagenized population to identify mutants unable to perform process of interest, affected gene required for &/or involved in that process, some types of screen: Mutagens used to increase frequency of finding mutations and thus the number of genes identified. Can change promoter enhancer sequence > change in expression. Screening start off with a large population and individually analyze each one of them for something. Go through each cell to assay its phenotype. More work than selection screening can be easy or laborious. It changes their morphology and their cell cycle. Once you"ve identified a bunch of mutants, now what? Phenotypic analysis: what exactly is altered in the mutant, morphology, biochem, behaviour.
