February 28, 2018: gtp binding to copii protein (sar1) doesn"t need gtpase activity, gtpase is needed to turn off the activity of the protein, hydrolysis of gtp allows the unfolding of the coat. Copi coated vesicles: retrograde transport (trans -> cis, golgi -> rer, the protein that is the gtpase is arf (or arf1) Soluble protein, binds to the kdel receptor: kdel (lys-asp-glu-leu, at the very c-terminus of the protein. Is the sequence of amino acids that signals to receptors in the golgi that the protein needs to be brought back to the rer: kkxx (lys-lys-x-x, membrane proteins, binds to copi. Inside is heavily glycosylated proteins to protect the rest of the cytoplasm from the hihgly acidic inside of the lysosome. Stomach of the cell, digest proteins and entire organelles (malfunctioning mitochondria: acid hydrolases: breaks down proteins at low ph only, proton pump (h+ atpase): part of the membrane that allow creation of a low ph environment.
