Can be accomplished by altering the activity of a protein or altering the amount of a protein. Protein amount can be altered at several levels. Rna polymerase starts transcription by binding to gene"s promoter. Additional proteins, called repressors and activators bind at or near the promoter and decrease or increase transcription. Specificity is achieved by recognition of a particular sequence. Sequences are typically inverted repeats (same sequence in opposite direction on opposite strand) Dna binding protein is a dimer (two subunits), each binding to the same sequence on opposite strands. Binding can either reduce transcription or increase. Reduced transcription -> lower transcript levels -> lower protein levels. Genes are negatively regulated in the presence of a particular molecule.
