This cyclic amp dependent- protein kinase which basically phosphorylates the calcium channel making it available for use: cell-atach patch used. Now, in this paricular case if they actually add the agonist in the bath they will see a response in the patch membrane. This a diagnosic test whether acivaion of the neurotransmiter receptor does actually leads to acivaion of second messenger. With the cell-atached patch situaion, if looking at ionotrophic receptor and add the nt into the bath, you are only acivaing the ion channel found outside of the patch, thus will not produce a response in the patch. Adding nt to the bath in the cell-atached patch is the diagnosic tool to determining whether difusible second messenger is produced or not.
