Bordetella pertussis is the causative agent of whooping cough: respiratory tract infection. 1998: 45 million cases, 400 000 deaths, even though inactivated vaccine was available, heat treated and killed with acetone, vaccine side effects, thus, purified vaccine developed. Site direction mutagenesis has been used to detoxify pertussis toxin: change toxin to toxoid.
