If no stain, can look at the refractive properties of tissue -> can look at interference and see what structures are causing this interference. If the refracted light is collected -> will produce some sort of image. Electron microscopes are limited can"t look at much other than where heavy metal stains are found. Looked at one light source at a time (one pixel at a time)
