Document Summary

Normally have significant impact on pathology and disease course. Could have subclinical disease way before symptoms appear. So when measuring autoantibodies we don"t know whether they are secondary, tertiary or quaternary to the event that triggered disease. If pathogenic they have impact on course of disease and manifestation. Auto-abs to gastric parietal cells binding to stomach in pernicious anemia. Lupus describes skin lesions evocative of a "wolf" bite. Chronic, often severe, autoimmune disease, sometimes lethal due to multi organ failure. Renal failure and vasculitis are major health problems in sle and leading causes of death. Immune complex deposition, inflammation, malar rash (butterfly-like rash on cheekbones), alopecia (hair loss), vascular damage, renal disorder, neurological disorder. Patients go from being very well to very bad. Not many good therapies: can be reasonably well managed but patient condition still deteriorates. Strong polygenic effect (shown by linkage analysis to cause susceptibility and also due to ethnicity), and also environmental influence.
