Characterization of the bcr was facilitated by the fact that the ig could also be secreted. Understanding the protein structure of the tcr was critical to the process of discovering the genes. Used monoclonal ab (mab) specific for tcr to purify and characterize it as an heterodimeric protein. Each chain has 1 variable and 1 constant region domain. Both chains contained regions arranged in characteristic ig fold. Presence of constant/variable regions provided clues as to potential arrangement of genes. The -chain genes was discovered simultaneously in two different laboratories. 1984, discovery of tcr genes in mice and humans by davis and hedrick. Davis-hedrick strategy based on 4 assumptions about tcr genes. Tcr genes will be expressed in t cells but not b cells. Since genes encode membrane-bound receptor, transcribed mrna will be found associated with membrane-bound polyribosomes. Genes will code for a variable and constant region. Genes will undergo rearrangement in t cells.
