Document Summary
Provide an overview of the pathogenesis of parkinson"s disease and parkinsonism. Assess genetic, environmental, and infectious risk factors for these illnesses in the context of primary and exacerbating causes. Parkinson"s disease (pd) (=primary parkinsons = idiopathic parkinsons= parkinson"s disease proper): A progressive neurological disease characterized by muscular tremor, deterioration of coordinated movement, partial facial paralysis, and weakness (often referred to as idiopathic pd ) Parkinsonisms (=secondary parkinsons = parkinsons attributable to a known cause): diseases with symptoms similar to pd that result from head injury, encephalitis, syphilis, carbon monoxide poisoning, cerebral arteriosclerosis, or use of mptp (mptp is a synthetic narcotic: 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine) Attributable to destruction of dopaminergic neurons in the substantia nigra. Lewy bodies containing synuclein are present in the damaged areas. 15-20% of afflicted have an afflicted family member. Rare form of pd shows a strong genetic basis; contursi family. -extended family in greece, italy & new jersey. -linked to a mutation in the synuclein gene.
