In this experiment, the main purpose is to familiarize with the use of various absorbance spectrometers and measure the protein concentration of two different unknowns by using biuret, lowry, In spectrometer orientation lab session, different instruments will be used to record the wavelength spectrum from the protein sample. In protein concentration lab session, by comparing the results from these methods, it is possible to deduce which samples were protein mixture, and which sample containing interfering substances. All the instructions were followed as the protocol handout provided by the teaching assistants on. The procedure followed is found on chapter 4 (page 116-118) of the laboratory textbook: For the first day of spectrometer orientation lab, we did not do the photometric part for hitachi and shimadzu"s spectrophotometer. All the reagents were obtained from the teaching assistants, we did not have to prepare the reagents for the experiment ahead of time.
