Mitochondrial dna tests do not individualize the evidence, but it does narrow down the possible source. You can use either 10x or 20x objective which will result in a magnification of 50 or 100 power. 5x would be too small and 45x would be too large. The perfect magnification to look at hair under a microscope is 25 to. Cuticle outermost layer of hair; made of keratin; is not a smooth layer but a series of overlapping scales like shingles on a roof. Cortex middle layer of hair; in human hair most thickest; made up of spindle-shaped cells and transparent; contains pigment granules, cortical fusi, and ovoid bodies. Medulla innermost layer of hair, consists of cells forming a shaft that in human hair can be either absent, fragmented or interrupted (continuous only in animal hair) There is a color difference between the tip and the shaft portion of the hair. The tip has a rood with a bulbous shape.
