Step 4: Aldolase
Breaks the ring ΔG’° = 23.8 kJ/mol works through a
complex multistep mechanism involving two Schiff bases;
only works when there is a carbonyl group on C2 & an
hydroxyl group on C4 necessary for the Schiff base
mechanism of action (it’s a lyase)
Makes Dihydroxyacetone phosphate (DHAP) &
Glyceraldehyde-3-phosphate (GAP)
Step 5: Triose Phosphate Isomerase (TIM/TPI)
- Converts DHAP GAP in a diffusion controlled process
- Mechanistically, its just His and Glu playing with a
couple of protons to catalyze an ene-diol
rearrangement can go both ways
Phase 2: The Payoff
ΔG°: -32.0 kJ/mol per molecule of glucose
- Step 6: Glyceraldehyde-3-phosphate dehydrogenase
