Document Summary

We have a patched protein (receptor) and smoothened protein. w/ out ligand (hedgehog), patch inhibits smoothen. And ci is degraded and you get no transcription. So not only do we not get transcription we also don"t get any repression. However in the presence of hedgehodg, it binds to patch, makes it unable to work so you lift repression (of patch onto smoothen) on smoothened. So smoothened is activated (in the presence of hedgehog) here and it protects ci from degredation (no need to know how), it then gets into the nucleus and turns on genes. Active smoothened inhibits a destruction complex which cleaves ci. Only need to memorize the box, dw about the rest. Wnt is important in many different signalling paths. Canonical(most important/premier pathway) is the one we need to understand. In the absence of wnt, beta catenin gets degraded. Similar to the hedge hog path(where ci is degraded).
