Document Summary

To characterize a white blood cell, these cells express a certain protein (transmembrane proteins, sticks out on the outside) Have an antibody that recognizes thy1 and another that recognizes cd3. Conjugate these antibodies to fluorophores and they will recognize proteins only on the outside. Can add dye that can cross plasma membrane and label dna. Antibodies made against specific cell surface proteins can be linked to fluorophores. Membrane permeable fluorescent dyes can be used to label intracellular structures (i. e. hoechst stain binds dna in nucleus). Cells with bound antibodies or that have taken up the dyes can now be sorted and counted. There are specific antibodies, which recognize the extra cellular domains. Hoechst stain (dye that can cross into the cell) has a high affinity for dna. Can show how much dna is in the cell. Lasers are illumination source, which are aimed at tubes.
