FSC402H5 Lecture Notes - Lecture 7: Streptococcal Pharyngitis, Immunoassay, Analytical Technique
Document Summary
Analytical technique based on chemistry & immunology. Body produces ab in response to foreign substance. Ab= designed to complex with specific drugs/drug classes. Ab have high specificity & affinity for specific antagonist: set-up, ab are attached/immobilized onto well, labels/tags (reagent, label- react as part of assay, so change in signal can be measured. Competitive format: blood sample containing analyte of interest (ag, labeled drug (ag*, both complete for immobilized ab, allow for equilibrium to occur, wash away excess reagent, measure signal from labeled drug (ag*) Immunoassay signal: more ag* attached to ab means greater signal, less ag in sample, enzyme-linked immunosorbent assay (elsia) Inverse relationship between drug (ag) concentration & signal intensity. Analytical overview for ft testing: volume of biological fluid: preselected volume by pipetting device, weight of solid & semi-solid tissue: must be homogenized with water/buffer prior to analysis.
