Glu1 transporter is responsible for basal glucose uptake in the muscle cell, it is not insulin responsive. Glu4 transporters are responsive to both insulin and muscle (?) Exercise does not induce phosphorylation of the insulin receptor or irs1. Aicar- and contraction-induced glucose transport are not inhibited by wortmannin. Aicar: ampk agonist, bypass the leptin receptors and directly activate ampk to the membrane and increase glucose uptake. At basal level, glu1 on the membranes take up glucose. With aicar: ampk activated, and increase glu4 translocation. Muscle contraction: activates ampk and glucose uptake increase through that pathway, so wortmannin does not have any effects on it. With insulin: glucose uptake increases; but decreases when pi3k is inhibited with wortmannin. Additive effects of aicar plus insulin and contraction plus insulin but not aicar plus.
