There is a direct correlation between stemness and dye efflux. Non-sc take up the dye, it intercalates with dna, lights up the cell. Flow cytometry to look for the dark or side population (the population that excludes the dye). Flow cytometry: there is a machine that sucks up cells one at a time. As the cells pass through the machine, there is a laser that shoots a beam. You can couple lasers in order to determine what markers are present or absent from the cell.
