Hepatocytes can store lipids and get rid of. Epithelial cells in mammary glands: push lipid droplets through pm (fat in milk), unconventional secretion pathway. Nucleus & some cytoplasm to side, most cell is one huge lipid droplet. Macrophages take up damaged ldl w/scavenger receptors foam cell die. Triglycerides has glycerol 3 fatty acids, no head group, not stable in er membrane triglycerides. & cholesterol ester form bulge in bilayer bud into cytoplasm as lipid droplet. Red: triglycerides, cluster in middle of bilayer bulge. Dgat2 can move to surface of lipid droplet. Acat stay in er, lipid droplet must associate w/er to add cholesterol ester. Dgat2 makes triglycerides on surface of lipid droplet, doesn"t involve er. Add oleic acid (fatty acid, 50% of molecule) dgat2 relocates to lipid droplets. Make triglycerides at lipid droplets lipid droplets get bigger. Lipid droplets will float easily on gradient, easily do membrane fractionation and purify. Lipid droplets can move a little when small.
