Lecture 13 – Generalized Anxiety Disorder
• Anxiolytics
1. Recall:
a. Act on the GABAA receptor
b. Proscribed for insomnia
c. Problem:
- Addiction potential
- Dangerous when mixed with alcohol
- Impair memory
d. Example:
- Benzodiazepine
- Valium
2. Newer version of Benzodiazepines:
a. Positive Allosteric Modulators:
- PAM:
i. Positive:
Activate GABAA receptor
ii. Allosteric:
Bind to the allosteric binding site
iii. Modulators:
Modulate themselves
- Three types:
i. Full allosteric modulators (most dangerous/highest abuse potential;
e.g., Xanax):
Act with high potency and efficacy on many GABAA receptors
ii. Selective allosteric modulator (e.g., Valium):
Act with high potency and efficacy but only on a select number of
the GABAA receptors
iii. Partial allosteric modulators (safest; e.g., Ambien):
Act with high potency but low efficacy
• Fear vs. Anxiety vs. Stress
1. Anxiety:
a. Thinking about going to the dentist
2. Fear:
a. Beig i the detist’s hair
3. Stress:
a. Our bodies response to something pushing against homeostasis
b. Allostatic load:
- It’s the oiatio of stress, poor diet, ad iatiity that akes stress
destructive and cause our modern-day illnesses
