Background and context

Fred Sanger invented the chain termination method of sequencing. This approach involves utilising a primer and a regular deoxynucleotide to replicate a single-stranded DNA template (dNTP). Labeled dideoxynucleotides are combined with the primer and dNTP (ddNTPs). Because the ddNTP lacks a  ' – OH group, it functions as a chain terminator, preventing more nucleotides from being added to the new chain.
